چکیده

Bleomycin-induced breakage of an isolated covalently closed circular DMA from bacteriophage PM2 was as sayed fluorometrically after agarose gel electrophoresis and staining with ethidium bromide. When bleomycindamaged DMA was assayed under neutral conditions, there was a decrease in the amount of Form I DMA and a simultaneous increase in both Forms II and III of DMA. However, when the damage was assayed under nondenaturing alkaline conditions, there was a greater decrease in the amount of Form I DMA and a corresponding in crease in both Forms II and III DNA compared with neutral conditions. Approximately one alkaline-labile site was formed for every single-strand break introduced. The rate of formation of Form III DNA was found to be approxi mately twice as fast when measured under alkaline con ditions compared with neutral conditions. Reaction of bleomycin-treated PM2 DNA with the Escherichia coli apurinic specific endonuclease IV demonstrated that the loss of Form I DNA was very similar to that observed with alkali treatment suggesting that the bleomycin-induced alkaline-labile damage is the result of base removal from the DNA helix. Similar results were obtained when the bleomycin-treated DNA was reacted with E. coli endonu clease III. However, no increase in double-strand break age was observed with either endonuclease III or IV, which may reflect an inability of the endonuclease to cleave at a site of base loss across from a single-strand break or another lost base. Alkali treatment of purified Form I DNA which was pretreated with bleomycin revealed that some of the alkali-induced double-strand breaks arose from Form I molecules, suggesting that two alka line-labile sites are formed across from each other. In addition, some Form II molecules produced by bleomycin treatment were converted to Form III molecules by nondenaturing alkaline conditions.

Influence of the antioxidant N-acetylcysteine and its metabolites on damage induced by bleomycin in PM2 bacteriophage DNA.

Bleomycin is considered to be a useful model compound for studying environmental carcinogenesis, due to its broad spectrum of DNA damaging properties. In addition, bleomycin is a useful antitumor drug because of its cytotoxic properties. To investigate the influence of the antioxidant N-acetylcysteine and its metabolites glutathione and cysteine on bleomycin-induced DNA damage and more importan...

Effects of DMA Superhelical Changes Induced by Ethidium Bromide on the DMA-degrading Activity of Two Antitumor Antibiotics, Bleomycin and Phleomycin1

The effects of changes in the conformational state of DNA on the single-strand and double-strand breakage activity of two antitumor antibiotics, bleomycin (BLM) A2 and phleomycin Di, have been studied by the gel electrophoretic analysis of the drugdegraded PM2 phage superhelical DNA pretreated with an inter calating agent, ethidium bromide (EB). Both the single-strand and double-strand breakage...

Breakage of a DNA-protein complex induced by bleomycin and their repair in cultured mouse fibroblasts.

The effect of bleomycin on DNA strand breakage and its repair in cultured mouse fibroblast strain L-P3 (a substrain of L929) was studied. Treatment of L-P3 cells with bleomycin caused single-strand breaks of DNA as revealed by alkaline sucrose gradient centrifugation. The singlestrand breaks of DNA were rejoined within 3 hr when the treated cells were incubated at 37°in a medium free of the an...

Damage and repair induced by bleomycin in the domain of human amplified MYC oncogenes.

Growing monolayer COLO320 human cells having a 30- to 40-fold amplification of a MYC domain were pulse treated for 15 min with increasing doses of bleomycin (BLM). Cellular DNA was extracted at the end of the BLM treatment or after an interval of 30 min, during which the cells were allowed to repair the DNA damage at 37 degrees C in culture medium without BLM. Damage and repair in total cellula...

Photo-induced reactions of benzo(a)pyrene with DNA in vitro.

The induction of lesions in DNA by combinations of benzo(a)pyrene (B(a)P) and near-ultraviolet light (NUV) with wavelengths of 300 to 480 nm was investigated. Using centrif ugal methods or agarose gel electrophoretic techniques to monitor the conversion of the superhelical double-stranded PM2 DNA to the relaxed circular form, it was possible to quantitate the rates of single-strand breakage in ...
